Cysteine Sulfhydryls in Tau Display Unusual Hyperreactivity

چکیده
Pathological tau aggregation is one of the major hallmarks Alzheimer's disease, for which there are no effective treatments thus far. We previously reported that cinnamaldehyde, in cinnamon, could inhibit vitro by reacting with or both cysteines tau. Herein, we have investigated reaction kinetics and found cinnamaldehyde displays significant hyperreactivity towards compared to model sulfhydryl compounds as well a short peptide corresponding sequence surrounding cysteines. Further, specific opposed other sulfhydryl-containing cellular proteins. Cinnamaldehyde several characteristics make it favorable potential inhibitor vivo: water lipid soluble, brain-permeable, non-toxic, inexpensive. The preferential targeting an additional property therapeutic lead compound.

Redox Factor-1 Activates Endothelial SIRTUIN1 through Reduction of Conserved Cysteine Sulfhydryls in Its Deacetylase Domain
Apurinic/Apyrmidinic Endonuclease 1/Redox Factor-1 (APE1/Ref-1) is a reductant which is important for vascular homeostasis. SIRTUIN1 (SIRT1) is a lysine deacetylase that also promotes endothelium-dependent vasorelaxation. We asked if APE1/Ref-1 governs the redox state and activity of SIRT1, and whether SIRT1 mediates the effect of APE1/Ref-1 on endothelium-dependent vascular function. APE1/Ref-...
متن کاملSulfhydryls-a new factor in frost resistance. II. Relation between sulfhydryls & relative resistance of 15 wheat varieties.
The wheat varieties were supplied by Dr. G. Aufhammer of the Institut fur Acker-und Pflanzenbau, at Freising-Weihenstephan, Germany. To obtain seedlings in the three-leaf stage preparatory to hardening, they were grown in flats of soil in the greenhouse undler prevailing temperatures during May to July. A photoperiod of 9 hours was used at all stages. The three-leaf seedlings were then hardened...
متن کاملAging and oxidation of reactive protein sulfhydryls.
Protein sulfhydryls are potential sites of reversible oxidative modification by S-glutathiolation, and S-nitrosylation, but they are also susceptible to irreversible damage by oxidative conditions. In the absence of adequate antioxidant protection, these reactive sites may become useless because of this irreversible damage. It has recently become possible to directly access the nature and amoun...
متن کاملJarring bodies: thoughts on the display of unusual anatomies.
This past April I woke up suddenly from a frightening yet laughable little dream in which I couldn’t breathe because Governor John Engler, dressed in a power suit, was sitting on my naked chest. My dreams have always been this transparent; they would bore a psychotherapist. I knew immediately when I awoke what this dream was about. The next day I had an appointment with a professional photograp...
متن کاملSulfhydryls and the in vitro polymerization of tubulin
The free sulfhydryls of brain tubulin prepared by cyclic polymerization procedures both with and without glycerol have been examined. The average free sulfhydryl titer of tubulin prepared with glycerol (7.0 sulfhydryls/55,000 mol wt) is greater than that of tubulin prepared without glycerol (4.0 sulfhydryls/55,000 mol wt). Diamide, a sulfhydryl-oxidizing agent, inhibits the polymerization of tu...
